Activity Overview
As the oral selective estrogen receptor degrader (SERD) class matures, attention is shifting from monotherapy to rational combination strategies intended to deepen and prolong disease control in hormone receptor–positive/HER2-negative (HR+/HER2–) metastatic breast cancer (MBC). Combining oral SERDs with CDK4/6 inhibitors or agents targeting the PI3K/AKT/mTOR (PAM) pathway provides a mechanistically grounded approach to overcoming endocrine resistance. However, these regimens also raise practical questions regarding patient selection, treatment sequencing, and additive toxicities.
In this activity, 2 experts examine the emerging evidence for oral SERD–based combinations. They review data about SERD–CDK4/6 inhibitor combinations, including evolving first-line paradigms and later-line umbrella trial results from EMBER-3 and ELEVATE. They also discuss SERD–PAM pathway strategies, including giredestrant plus everolimus (evERA) and capivasertib- and gedatolisib-based combinations. Finally, the faculty explore how to incorporate these data into individualized treatment decisions and outline evidence-based approaches to anticipating, monitoring, and managing adverse events associated with targeted combination regimens.
Target Audience
This educational program is primarily directed toward a global audience of medical oncologists, advanced practice providers, nurses, and other health care professionals who are involved in the management of patients with advanced breast cancer or MBC.
